Yes, there were a couple of crashing bugs that made it into 1.0.0 and were fixed in 1.0.1, but the change log only says "crash fixes", it doesn't say specifically what, so let us know how that works out.

You can ad them to ops.txt or you can go to the Server control tab, open the console and use the command "op NAME" where NAME is the name of the person you want to op.

Bukkit plugins generally just need to be uploaded to the "plugins" folder in the server FTP, once Bukkit has been installed. Some plugins have more complicated configurations, and they should either talk about this in their forum post or in a readme file.
